A method used with electron microscopy (EM), which is basically freezing the specimen very rapidly, vacuuming off the ice crystals which, because frozen rapidly, are loose and disorganized, spraying with metal vapor (metal is used because it provides significant contrast in EM) and then using acids to dissolve away the organic matter. So what you are left with is a thin metal shell that is in the shape of the cell.
