In a martial arts context, to chamber is to set the body part used to strike in a convinient ready position. This position is intended to create the optimum conditions to strike from. The chambering position should create the most tension in the muscles used to strike as well as the shortest distance from the ready position to the spot being hit. The chambering position is generally integral to any strike because it generates the most power and speed.
